When I do close ups of eyes I like to add more details like veins and stuff. If I show the inside of a character's mouth, I try to draw the different teeth, delineating between incisors, canines and molars. I try to show the difference between Isis and Rahab's hands...

Talking about a character's personality is a good indicator that they have one at all. I've seen many comics where the protagonist doesn't even have enough personality for the creator to discuss at all, they just talk about the art. Character depth shows creator passion.
