ClayBuildup is great for sculpting, but it can also be really great for polypainting! By selecting a darker colour than the base colour, setting the brush to drag rectangle and removing the alpha (optional?), you can apply some really nice colour variation and gradients.

I always start with a paper sketch, then jump on Illustrator, getting the main shapes right! Then i start working the gradient and colors, and i get to the point that i want. After this, After Effects and start animating :)
