Ryoko's involvement and the Mendo Family using the cast as pieces in their parcheesy game is actually borrowed from the original anime's adaptation. In the manga it's only planned by Mendo's parents, and the game is more of a meta-joke for readers to print out and play themselves
Both of these stories once again originally involved Ten, who has been written around. The adaptation of 121 gets around this by having Ataru and Lum walk to school together instead of Lum and Ten flying, their observations about the oxcart being made by Kosuke and Hokuto instead

Finally, the episode ends with a teaser for next week's Parent's Day episode, featuring the return of Lum's original seiyuu Fumi Hirano as Lum's mom, reuniting with Ataru's original seiyuu Toshio Furukawa as Ataru's Dad! 🤩😍✨
The only other notable anime addition is Lum yelling at Rei not to never come back again when he leaves the class, saving her line about being tired for after the added scene of her and Ataru coming home to find Rei being fed by Ataru's mom.
In the manga, Lum intentionally pushes Ataru's mom in Rei's way, whereas in the anime they she and Ataru run upstairs before Ataru's mom comes at the door as Rei arrives. I like Rei's proposal line better in the manga, since it's more to the heart (or stomach) of what he wants.😂
Lum and Ataru teasing Rei, saying she'll go back with him if his proposal's eloquent and mocking him for believing in her promise, is cut from the manga. Instead, Lum lovingly embracing Ataru is what enrages Rei into chasing them, which she's less amused about than in the manga
Rei breaking out his Lum doll and piggy bank marks the transition from chapter 6 content to chapter 7 content in the episode. In the manga, it's Ataru who goades Rei into proposing, which prompts him to break out the cheat sheet, whereas in the anime it was Rei's plan to propose.
